Comparative assessment of the bacterial communities associated with Anopheles darlingi immature stages and their breeding sites in the Brazilian Amazon

Background: The neotropical anopheline mosquito Anopheles darlingi is a major malaria vector in the Americas. Studies on mosquito-associated microbiota have shown that symbiotic bacteria play a major role in host biology. Mosquitoes acquire and transmit microorganisms over their life cycle. Specifically, the microbiota of immature forms is largely acquired from their aquatic environment. Therefore

The human cationic antimicrobial protein (hCAP18), a peptide antibiotic, is widely expressed in human squamous epithelia and colocalizes with interleukin-6

Peptide antibiotics are widespread in nature and, by providing a rapid first line of defense, may be key players in the innate immune system. Although epithelia are the main barriers shielding the internal environment from microorganisms, the role for peptide antibiotics in epithelial protection is unclear. We recently reported that the human cationic antimicrobial protein hCAP18, the precursor of

ARID1a Associates with Lymphoid-Restricted Transcription Factors and Has an Essential Role in T Cell Development

Maturation of lymphoid cells is controlled by the action of stage and lineage-restricted transcription factors working in concert with the general transcription and chromatin remodeling machinery to regulate gene expression. To better understand this functional interplay, we used Biotin Identification in human embryonic kidney cells to identify proximity interaction partners for GATA3, TCF7 (TCF1)

Recognition of maturity-onset diabetes of the young in China

Aims/Introduction: Given that mutations related to maturity-onset diabetes of the young (MODY) are rarely found in Chinese populations, we aim to characterize the mutation spectrum of MODY pedigrees. Materials and Methods: Maturity-onset diabetes of the young candidate gene- or exome-targeted capture sequencing was carried out in 76 probands from unrelated families fulfilling the clinical diagnost

Decreased HLA-DQ expression on peripheral blood cells in children with varying number of beta cell autoantibodies

The risk for type 1 diabetes is strongly associated with HLA-DQ and the appearance of beta cell autoantibodies against either insulin, glutamate decarboxylase (GAD65), insulinoma-associated protein-2 (IA-2), or zinc transporter 8 (ZnT8). Prolonged exposure to autoantibodies may be related to T cell exhaustion known to occur in chronic infections or autoimmune disorders. It was hypothesized that au

In between resolution and cutting planes : A study of proof systems for pseudo-boolean SAT solving

We initiate a proof complexity theoretic study of subsystems of cutting planes (CP) modelling proof search in conflict-driven pseudo-Boolean (PB) solvers. These algorithms combine restrictions such as that addition of constraints should always cancel a variable and/or that so-called saturation is used instead of division. It is known that on CNF inputs cutting planes with cancelling addition and s

Glucose-6-phosphate dehydrogenase deficiency genotypes and allele frequencies in the Kavango and Zambezi regions of northern Namibia

Background: Namibia has made significant gains in the fight against malaria, with a target of elimination by 2023. We examined the genotype and allele frequencies of glucose-6-phosphate dehydrogenase (G6PD) deficiency to inform decisions on primaquine use, as we recently detected clusters of Plasmodium ovale curtisi in Kavango. Methods: A multistaged cross-sectional sampling method was used to enr

Undifferentiated pleomorphic sarcomas with PRDM10 fusions have a distinct gene expression profile

Undifferentiated pleomorphic sarcoma (UPS) is a highly aggressive soft tissue tumor. A subset of UPS is characterized by a CITED2–PRDM10 or a MED12–PRDM10 gene fusion. Preliminary data suggest that these so-called PRDM10-rearranged tumors (PRT) are clinically more indolent than classical high-grade UPS, and hence important to recognize. Here, we assessed the spectrum of accompanying mutations and

Cadmium, carotid atherosclerosis, and incidence of ischemic stroke

Background--Exposure to cadmium has been associated with carotid plaques, inflammation in carotid plaques, and increased risk of ischemic stroke. This study examined the separate and interacting effects of blood cadmium levels and carotid plaques on the risk of incident ischemic stroke. Methods and Results--Cadmium levels were measured in 4156 subjects (39.2% men; mean ± SD age 57.3 ± 5.9 years) w

Patentability of human enhancement : From ethical dilemmas to legal (un)certainty

Emerging technologies are paving the way for future revolutionary advances in science that may open the possibility to change the very anthropological definition of human being. This mere possibility has lead to ethical interrogations concerning the nature and boundaries of human nature and our relationship with science and technology. Meanwhile the Law has faced the challenge of reflecting on the

Aggregation potency and proinflammatory effects of SARS-CoV-2 proteins

Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, is primarily known as a respiratory disease. The continued study of the disease has shown that long-term COVID-19 symptoms include persisting effects of the virus on the brain when the infection is over, possibly even leading to neurodegeneration. However, the exact mechanisms of

Sex-Specific Cardiovascular Consequences of Long-Term High-Salt Diet in Mice

BACKGROUND: Excessive dietary salt intake is associated with elevated blood pressure and damage to organs including the heart, vasculature, and kidneys. Women are more prone to salt-sensitive hypertension, yet preclinical studies often focus on men.METHODS: To examine sex-specific responses to chronic salt loading, male and female C57Bl6/J mice were fed a high-salt diet (HSD; 8% NaCl) from 7 to 14

Speaker’s comfort and vocal effort in different room acoustic conditions : A controlled field experiment in a university lecture room

Speaking in poor acoustic conditions constitutes a risk factor for voice disorders. This study investigated how speaker’s comfort and vocal effort were affected during short speech tasks in four different room acoustic configurations. Voice recordings of the speech tasks and self-evaluations of the speaker’s comfort were collected from 15 participants in four room acoustic configurations as part o

Computed tomography grey-white matter ratio at the basal ganglia level in a reference population for hypoxic ischaemic encephalopathy after cardiac arrest – A cross-sectional observational study

Introduction: A grey-white matter ratio (GWR) < 1.10 at the basal ganglia level on head computed tomography (CT) previously predicted poor outcome with high specificity and moderate sensitivity in cardiac arrest patients. Data on GWR in reference populations are lacking. We explored GWR in a reference group to a cardiac arrest population and hypothesised that none would have GWR < 1.10. Methods: A
